An Improved Synthesis of 1,4,7-Triazacyclononanes (tacns) and 1,4,7,10-Tetraazacyclododecanes (cyclens)

Reaction of tosylbis[2-(tosyloxy)ethyl]amine with ethylenediamine gives 1-tosyl-1,4,7-triazacyclononane in good yield. A similar reaction of tosylbis[2-(tosyloxy)ethyl]amine with diethylenetriamine gives 1-tosyl-1,4,7,10-tetraazacyclododecane. These tosylated compounds can be further transformed to give 1,4,7-triazacyclononane (tacn) or 1,4,7,10-tetraazacyclododecane (cyclen) or their derivatives
甲苯磺酰基双[2-(甲苯磺酰氧基)乙基]胺与乙二胺的反应得到1-甲苯磺酰基-1,4,7-三氮杂环壬烷,收率很高。甲苯磺酰基双[2-(甲苯磺酰氧基)乙基]胺与二亚乙基三胺的类似反应得到1-甲苯磺酰基-1,4,7,10-四氮杂环十二烷。这些甲苯磺酸化的化合物可进一步转化为1,4,7-三氮杂环壬烷(tacn)或1,4,7,10-四氮杂环十二烷(环素)或其衍生物。 环胺-环化-脱甲苯基化-大环-甲基化
