A phosphorothioate diester can be incorporated site-specifically into DNA sequences to provide a nucleophilic site that is amenable to alkylation by labels containing haloacetamide, aziridine sulfonamide, or γ-bromo-α,β-unsaturated carbonyl functionalities.
