A method of treating cancer, inflammatory disease or an infectious disease or condition in a subject in need of such treatment is disclosed. The method comprises administering to a subject an effective amount of a manzamine, or a rationally modified manzamine derivative or analog or an optical isomer or racemate or tautomer thereof or a pharmaceutically acceptable salt or prodrug thereof generated through optimized fermentation of a

described. The key step, formation of the 1-phenylacetyl β- carboline, involves a tandem aza Wittig / electrocyclicringclosure process. The first synthesis of the alkaloid xestomanzamine A is achieved by coupling of a N-protected harmane, now available via aza Wittig / electrocyclicringclosure process, with a 5-lithioimidazole derivative.
Two new β-carboline alkaloids, xestomanzamine A (1) and xestomanzamine B (2), and a new manzamine-type alkaloid, manzamine X (3), were isolated as cytotoxic constituents from an Okinawan marine sponge of Xestospongia sp. Another new manzamine-type alkaloid, manzamine Y(4), was isolated from an Okinawan marine sponge of Haliclona sp. The structures of these alkaloids were elucidated on the bases of
