Abstract The title compounds 4 have been obtained in high yields in a one-step sequence by the Mitsunobu reaction of diethyl 1-hydroxyalkylphosphonates 1 with hydrazoic acid, and subsequence treatment of the inter-mediate azides 2 with triphenylphosphine, followed by hydrolysis of the iminophosphoranes 3 with water.

Diethyl 1-azidoalkylphosphonates were obtained in high yield, and under mild conditions by the reaction of diethyl 1-hydroxyalkyl-phosphonates with hydrazoic acid in the presence of triphenylphosphine/diethyl azodicarboxylate (the Mitsunobu reaction).

for the synthesis of protected phosphonamidate peptides from O-ethyl 1-azidoalkylphosphonic acids and amino acids esters was developed. The method is general and the corresponding N-Boc protected phosphonopeptides as well as their azido analogs have been obtained in good yields. An efficient preparation of O-ethyl 1-azidoalkylphosphonic acids is also presented.

Abstract A simple and safe method for the preparation of dialkyl 1‐azidoalkylphosphonates from dialkyl 1‐(4‐nitrobenzenesulfonyloxy)alkylphosphonates (dialkyl 1‐(nosyloxy)alkylphosphonates) and tetramethylguanidinium azide (TMGA) has been developed.
