Process for preparation of co-crystal of lamivudine with zidovudine comprising of trituration of equimolar quantities of Lamivudine and zidovudine in presence of water at ambient temperature, optionally adding organic solvent during trituration, and drying the product. Process for preparation of co-crystal of lamivudine with zidovudine comprising stirring equimolar quantities of lamivudine and zidovudine in an organic solvent in presence of water at ambient temperature, evaporating solvents of the solution, allowing crystallization of product with optional seeding, isolating co-crystals and drying. Pharmaceutical composition comprising therapeutically effective amount of co-crystals of lamivudine, zidovudine.
