Highly enantioselective desymmetrizations of meso-anhydrides

Readily available, low molecular cyclohexane-based organocatalysts promote highlyenantioselectivedesymmetrizations of cyclic meso-anhydrides applying alcohols and benzyl mercaptan as nucleophiles. Both succinic and glutaric anhydrides furnished the corresponding products with up to 96% ee in mostly quantitative yields.

본 발명은 (R,R)-1,2-다이페닐에틸렌다이아민(DPEN) 기반의 티오우레아 촉매 화합물을 이용한 키랄 카르보닐 유도체의 제조방법 등에 관한 것으로서, 상기 제조방법에 따르면 고리형 무수물과 티올(-SH) 또는 아민(NH2)의 비대칭 고리열림반응, 즉 황 분해반응(thiolysis) 또는 질소 분해반응(aminolysis)를 통해 높은 거울상선택성을 가진 키랄 카르보닐 유도체를 우수한 수율로 제조할 수 있다.
Highly Enantioselective Thiolysis of Prochiral Cyclic Anhydrides Catalyzed by Amino Alcohol Bifunctional Organocatalysts and Its Application to the Synthesis of Pregabalin
Asymmetric thiolysis of prochiralcyclicanhydrides was achieved with our developed chiralsulfonamide and squaramide bifunctional organocatalysts based on amino alcohol scaffolds. The corresponding thioesters were obtained in high yields with excellent enantioselectivities. The usefulness of this methodology was demonstrated in the enantioselective synthesis of pregabalin.
