  Inhibitors of the Interaction of a Thyroid Hormone Receptor and Coactivators:  Preliminary Structure−Activity Relationships

  摘要：

  The modulation of gene regulation by blocking the interaction between the thyroid receptor (TR) and obligate coregulators has been reported recently with the discovery of the lead compound 3-(dimethylamino)-1-(4-hexylphenyl)propan-1-one). Herein we report studies aimed at optimization of this initial hit to determine the basic parameters of the structure -activity relationships and clarify the mechanism of action. These studies provided new insights, showing that activity and TR beta isoform selectivity is highly correlated with the structural composition of these covalent inhibitors.

  The ability of aqueous micelles and reversed micelles to control the regiochemistry of Diels-Alder reactions of (E,E)-6-[[[[4-[(4-hexylphenyl)sulfonyl]-1,3-butadienyl]amino] carbonyl]oxy]-N,N,N-trimethyl-1-hexanaminium bromide (4) and 1-(4-hexylphenyl)-2-propen-1-one (2b) was evaluated. If 2b and 4 were to react within the micelles in their preferred orientations, cycloadduct 10 would result, as opposed to 6-[[[[cis-6-(4-hexylbenzoyl)-cis-4-[(4-hexylphenyl)sulfonyl]-2-cyclohexen-1-yl]amino]carbonyl]oxy]-N,N,N-trimethyl-1-hexanaminium bromide (7b) and its exo isomer 8b, the theoretically predicted products actually obtained. The orientational effects in the aggregates were not strong enough to overcome the reaction's intrinsically preferred regiochemistry.

  A series of quaternary alkylammonium sulfobetaines of general formula RN+(CH3)2(CH2)nSO3 -, where n = 2-4, have been synthesised by reacting the corresponding N,N-dimethylamines with either sodium 2-chloroethanesulfonate (n = 2), 1,3-propanesultone (n = 3), or 1,4-butanesultone (n = 4). Full details of the preparation of the required N,N-dimethylamines are reported.

  A series of 3,6-substituted cyclohex-2-en-l-ones have been prepared by an efficient convergent Robinson-type annulation route. The cyclohex-2-en-l-one ring is the basis of a novel mesogenic core and when substituted at the 6-position provides a chiral centre, which is adjacent to a strong transverse dipole moment. This system has the advantage that both of the two main features required for new electro-optical applications (a dipole for switching and a chiral centre to reduce the symmetry) are located in the molecular core. The synthesis and mesogenic properties of these novel enones are reported.

  Die Erfindung betrifft ein Verfahren zur Diagnose einer genetischen Prädisposition für Brustkrebs (Mammakarzinom) und/oder Eierstockkrebs (Ovarialkarzinom) eines menschlichen Individuums, das dadurch gekennzeichnet ist, dass man in einer Probe des Mammagewebes und/oder Eierstockgewebes des Individuums die Konzentration der Thyroidhormonrezeptoren alpha und beta (THRa und THRß) bestimmt, wobei eine erhöhte Konzentration an THRa und/oder THRß verglichen mit gesundem Gewebe ein erhöhtes Risiko des Individuums für Brustkrebs und/oder Eierstockkrebs anzeigt sowie ein Verfahren zur Prognose der Schwere des Verlaufs eines primären Mammakarzinoms und/oder eines primären Ovarialkarzinoms eines menschlichen Individuums, das dadurch gekennzeichnet ist, dass man in einer Probe des Mammagewebes und/oder Eierstockgewebes des Individuums die Konzentration der Thyroidhormonrezeptoren alpha und beta ((THRa und THRß) bestimmt, wobei eine erhöhte Konzentration an THRa und/oder THRß verglichen mit gesundem Gewebe ein höheres Grading und einen ungünstigen Verlauf der Erkrankung anzeigt. Ferner betrifft die Erfindung ein Kit, umfassend a) ein gegen den Thyroidhormonrezeptor alpha/beta gerichtetes detektierbares Bindungsmolekül, b) Reagenzien und/oder Puffer zur qualitativen und/oder quantitativen Bestimmung des Thyroidhormonrezeptor alpha/beta-Bindungsmolekül-Komplexes, und c) Instruktionen zur Durchführung und Auswertung des Bestimmungsverfahrens, zur Verwendung zur Diagnose einer genetischen Prädisposition für Brustkrebs (Mammakarzinom) und/oder Eierstockkrebs (Ovarialkarzinom) eines menschlichen Individuums sowie ein therapeutisch wirksame Menge einer Verbindung die in der Lage ist, die Expression oder Aktivität des Thyroidhormonrezeptors alpha und/oder beta zu modulieren, zur Verwendung zur Propylaxe und/oder zur Behandlung von Brustkrebs und/oder Eierstockkrebs.
