    Extensive SAR and Computational Studies of 3-{4-[(Benzylmethylamino)methyl]phenyl}-6,7-dimethoxy-2H-2-chromenone (AP2238) Derivatives
    摘要:
    AP2238 was the first compound published to bind both anionic sites of the human acetylcholinesterase, allowing the simultaneous inhibition of the catalytic and the amyloid-beta pro-aggregating activities of AChE. Here we attempted to derive a comprehensive structure-activity relationship picture for this molecule, affording 28 derivatives for which AChE and BChE inhibitory activities were evaluated. Selected compounds were also tested for their ability to prevent the AChE-induced A beta-aggregation. Moreover, docking simulations and molecular orbital calculations were performed.

    A series of photodecarboxylative benzylations of N-methoxyphthalimide were successfully realised using easily accessible starting materials. The reactions proceeded smoothly and the corresponding benzylated hydroxyphthalimidines were obtained in moderate to good yields of 52–73 %. No competing photoinduced dealkoxylation of the N-methoxy group was observed. The reaction with potassium phenylacetate was subsequently investigated in an advanced continuous-flow photoreactor. The reactor allowed rapid optimization of the reaction conditions and gave the desired benzylated product in higher yield and shorter irradiation time compared with the batch process.

    A polyphenylene sulfide resin composition which comprises 100 parts by weight of a polyphenylene sulfide resin (A) and, compounded therewith, 1 to 100 parts by weight of glass fibers (B) having a single-fiber diameter of 12 μm or lager and has a crystallization temperature during cooling of 205° C. or lower. Also provided is a polyphenylene sulfide resin composition which comprises 100 parts by weight of a polyphenylene sulfide resin (A) and, compounded therewith, 1 to 100 parts by weight of glass fibers (B) having a single-fiber diameter of 10 to 12 μm, excluding 12 μm, has a crystallization temperature during cooling of 205° C. or lower and a chloroform-extractable content of 0.5 wt. % or lower, and gives a 2 mm-thick molded product which has a transmittance of 15% or higher for laser beam having a wavelength of 940 nm and a heat distortion temperature of 230° C. or higher under a load of 1.82 MPa.

    Aromatic sulfide/sulfone polymers are prepared by initially contacting a dihaloaromatic sulfone, an alkali metal sulfide, an organic amide, and, optionally, an alkali metal carboxylate under polymerization conditions and subsequently at or near completion of the polymerization reaction, adding an end-capping agent comprising additional dihaloaromatic sulfone to the reaction mixture producing a polymer exhibiting improved physical properties including melt flow stability. The resulting polymers have utility as coatings, films, molded objects, filaments, fibers, and the like.

    A method for producing arylene sulfide polymer in which a reaction mixture containing at least one polyhalo-substituted aromatic compound, at least one organic amide, at least one alkali metal carboxylate, H2S and water in an amount up to 5 weight percent of the total of the reaction mixture components is contacted at polymerization conditions for a time sufficient to produce polymer. In a preferred embodiment, at least one alkali metal carboxylate is present in the reaction mixture.
