    Unusual large Stokes shift and solvatochromic fluorophore: Synthesis, spectra, and solvent effect of 6-substituted 2,3-naphthalimide

    In this article four new series of 6-substituted 2 3-naphthalimides (1a-d 2a-d 3a-d 4a-d and 4 e-h) have been designed and synthesized through the formation of key cyclic anhydride intermediate which was the precursor of the well known environment-sensitive fluorophore [6-N,N-dimethylammonaphthalimides (6-DMN)] and other 6-substituted 2 3-naphthalimide series (2 3 4) Based on 6-amino-2 3-naphthalimide (6-ANP) compound a new type of fluorophore was found to exhibit moderate to unusual large Stokes shift (297-303 nm) 6-ANP derivatives display relatively low fluorescence quantum yields in high polar protic solvents such as water (Phi(F) similar to 0 004 571-576 nm) and a significant unusual red shift due to (1) hydrogen bonding interaction of the excited state of the molecule with the solvents which presumably enhance the intersystem crossing process in the system to quench fluorescence (2) this large Stokes shift was assumed to be the consequence of a substantial change of the geometric structure from the ground state (S-0) to the first excited state (S-1)) Compared with the other compounds studied the fluorescence of the nitro- and halo-derivatives was rather weak probably due to the efficient intersystem crossing leading to a non-reactive triplet state (C) 2010 Elsevier B V All rights reserved

    Sensors for detecting enzyme activity are provided. The sensors include substrate modules having environmentally sensitive labels and detection modules whose binding to the substrate modules results in changes in signals from the environmentally sensitive labels or polypeptides or polypeptide substrates including environmentally sensitive or fluorescent labels. Compositions including substrate modules, polypeptides, or polypeptide substrates and nucleic acids encoding enzymes and/or detection modules are also described. Methods of assaying enzyme activity using sensors including environmentally sensitive or fluorescent labels are provided, as are related methods for screening for modulators of enzyme activity.

    Provide is a method for detecting a target substance, which method can visualize the expression of the target substance at any time point while reducing influences on the functions of the target substance, can use fluorescent dyes having various excitation/emission wavelengths, and can achieve easy staining process. Also provided are a tag, a DNA, a vector, a probe and a detection kit suitable for use in the above-described detection method. Specifically, the method for detecting a target substance, comprises the steps of bringing into contact with each other (a) a tag comprising a polypeptide forming an α-helix structure, the tag bound to the target substance, and (b) a probe comprising a compound bound to a fluorescent dye; and measuring the fluorescence emitted by the fluorescent dye. The binding of the tag α-helix structure to the probe compound induces a spectral change in the fluorescence emitted by the fluorescent dye.

    The present invention generally relates to environment-sensitive fluorophores, including environment-sensitive fluorophores for reporting protein/protein and peptide/protein interactions. In one aspect, the present invention is directed to compounds and salts thereof, compositions and methods useful in determining biological interactions. In some cases, the compounds of the present invention are environment-sensitive fluorophores that have spectroscopic behavior that may depend on factors such as the physicochemical properties of the surrounding environment. The compounds of the present invention can be used, in certain embodiments, to monitor ions, small molecules, and biological processes such as protein folding, protein-protein interactions and phosphorylation events.
