  摘要：

  令人信服的是，定期服用阿司匹林可以降低结直肠癌的风险。但是，长期服用阿司匹林会导致胃毒性。本文中，我们设计和合成了一类新的基于白藜芦醇的阿司匹林前药，以同时释放阿司匹林和白藜芦醇以减轻由阿司匹林引起的副作用。前药RAH发挥了增强的抗癌活性，优于阿司匹林和白藜芦醇的物理混合物以及各自的抗癌活性。小鼠中RAH的代谢表明，大部分RAH在肠道内或吸收后被分解释放白藜芦醇和阿司匹林或水杨酸。机理研究表明，RAH通过下调细胞周期蛋白抑制细胞周期停滞，并通过激活癌细胞中的caspase-3诱导细胞凋亡。这些发现突显了基于白藜芦醇的阿司匹林前药的抗癌特性得到改善。RAH可能代表阿司匹林的新颖和安全替代品，以供日后日后使用。
