Expeditious Route Towards (±)-Desethyleburnamonine, a Precursor of (±)-Vindeburnol

An expeditious route to (±)-desethyleburnamonine (1), a direct precursor of vindeburnol, is reported. Microwave irradiation of an indolo-tetrahydropyridine ethyl ester 2 in CH3CN in the presence of 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) gave the pentacyclic (±)-desethyleburnamonine in a one-pot process. The invention is concerned with the compounds of formula (I) and pharmaceutically acceptable salts thereof. In addition, the present invention relates to methods of manufacturing and using the compounds of formula (I) as well as pharmaceutical compositions containing such compounds. The compounds of formula (I) are LMP7 inhibitors and may be useful in treating associated inflammatory diseases and disorders such as, for example, rheumatoid arthritis, lupus and irritable bowel disease.
