Design, synthesis, and biological evaluation of novel diarylalkyl amides as TRPV1 antagonists
摘要:
We have developed a new class of diarylalkyl amides as novel TRPV1 antagonists. They exhibited potent Ca-45(2+) uptake inhibitions in rat DRG neuron. In particular, the amide 59 was identified as a potent antagonist with IC50 of 57 nM. The synthesis and structure-activity relationship of the diarylalkyl amides are also described. Thermodynamic dissociation constants in various water-organic solvent mixtures are given for benzylammonium, benzyldialkylammonium, and (2-phenylethyl)ammonium ions. Deviations from the Hammett equation (Eqn. 1) are similar to those observed for carboxylic acids, but of opposite sign. The extended Hammett equation (Eqn. 3), containing the hydrophobic constant, π, yields good correlations. Derived secondary
Described are certain phosphite-isocyanurate oligomers and their use as thermal stabilizers in polymer compositions. The phosphite-isocyanurates may be prepared from a tris-hydroxyalkyl isocyanurate by reaction with an organic phosphite such as triphenyl phosphite or tributyl phosphite. Optionally, an alcohol such as decyl alcohol may be employed as a reactant.
