    Highly Sensitive Fluorescent Probes for Zinc Ion Based on Triazolyl-Containing Tetradentate Coordination Motifs
    摘要:
    Two new 1,2,3-triazolyl-containing N4-tetradentate ligands show nanomolar affinity for Zn2+ under physiological conditions. Furthermore, they are easily derivatizable to afford fluorescent probes suitable for sensitive Zn2+ detection. Structural characterizations in both solid and solution states suggest that N3 and N2 in the 1,2,3-triazolyl moiety coordinate with Zn2+ in A and B to afford five-membered and six-membered coordination rings, respectively. The probes show sensitive fluorescence enhancement and ratiometric responses to Zn2+.

    Direct Modulation of Microtubule Stability Contributes to Anthracene General Anesthesia
    摘要:
    Recently, we identified 1-aminoanthracene as a fluorescent general anesthetic. To investigate the mechanism of action, a photoactive analogue, 1-azidoanthracene, was synthesized. Administration of 1-azidoanthracene to albino stage 40-47 tadpoles was found to immobilize animals upon near-UV irradiation of the forebrain region. The immobilization was often reversible, but it was characterized by a longer duration consistent with covalent attachment of the ligand to functionally important targets. IEF/SDS-PAGE examination of irradiated tadpole brain homogenate revealed labeled protein, identified by mass spectrometry as beta-tubulin. In vitro assays with aminoanthracene-cross-linked tubulin indicated inhibition of microtubule polymerization, similar to colchicine. Tandem mass spectrometry confirmed anthracene binding near the colchicine site. Stage 40-47 tadpoles were also incubated 1 h with microtubule stabilizing agents, epothilone D or discodermolide, followed by dosing with 1-aminoanthracene. The effective concentration of 1-aminoanthracene required to immobilize the tadpoles was significantly increased in the presence of either microtubule stabilizing agent. Epothilone D similarly mitigated the effects of a clinical neurosteroid general anesthetic, allopregnanolone, believed to occupy the colchicine site in tubulin. We conclude that neuronal microtubules are "on-pathway" targets for anthracene general anesthetics and may also represent functional targets for some neurosteroid general anesthetics.

    This invention provides a process which comprises contacting, in a reaction zone, at least one organic azide, at least one alkyne, and at least one N-heterocyclic carbene copper compound in which the ligands are either (i) a halide and an N-heterocyclic carbene or (ii) two N-heterocyclic carbenes and a BF 4 − or PF 6 − anion, to form a 1,2,3-triazole in which at least the 1 and 4 positions each has a substituent. The N-heterocyclic carbene either an imidazol-2-ylidene in which the 1 and the 3 positions each has a substituent which has at least one carbon atom, or a 4,5-dihydro-imidazol-2-ylidene in which the 1 and the 3 positions each has a substituent which has at least one carbon atom.

    The present invention relates to π-conjugated fluoroionophores, methods for their preparation, and their use and a method for determining an alkali ion. Fluoroionophoric compounds of the general formula I are described Ionophore – π-Linker - Fluorophore (I) wherein the Ionophore is an anilino containing crown ether or cryptand with one or more anilino donor moieties as electron donors, forming a stable complex with an alkali metal ion the π-Linker is an aromatic or heteroaromatic conjugative linking moiety, and the Fluorophore is an electron acceptor moiety. Variation of the ionophoric unit offers a broad spectrum of detectable K+ and Na+ -concentrations, ranging from high concentration around 800 mM down to very low concentrations around 3 mM. The fluoroionophores have great potential for application in fluorescent optode system based blood analyzing equipment for methods and kits for the determination of K+ and Na+ concentrations in biological systems, either in vitro or in vivo, using embodiments of the disclosed fluoroionophores.
