    A spectrophotometric method has been developed for the determination of [(2-chlorophenyl)methylene]propane dinitrile (CS) stabilized by the diazonium salts Fast Red TR and Fast Blue B after extraction with chloroform. Mass spectrometry, 1H and 13C NMR and elemental analysis confirmed the presence of the hydrazoform of the azo dye formed by reaction with malononitrile, as a product of the alkaline hydrolysis of substance CS, and the diazonium salt Fast Red TR.

    The present invention provides an extremely simple and effective method of a sandwich EIA (enzyme immunoassay) system, or more precisely, a method for the detection of an antigen or an antibody by means of a sandwich enzyme immunoassay system using a combined material of insoluble carrier/antigen/enzyme-labeled antibody or a combined material of insoluble carrier/antigen/enzyme-labeled anti-immunoglobulin-antibody which comprises an insoluble carrier having coated thereon a layer of an antibody or antigen specific to the antigen or antibody to be detected and a layer of an enzyme-labeled antibody or enzyme-labeled anto- immunoglobulin-antibody specific to the antigen or antibody to be detected. According to the method of the present invention, the immunoreaction product comprising fixed antibody/ antigen to be detected/labeled antibody or fixed antigen/ antibody to be detected/labeled antiimmunoglobulin- antibody can be formed on the surface ofthe insoluble carrier by only one incubation and only one washing operation.

    An easy operable method of detecting phosphatase which comprises a step for the production of a dye to obtain an azo dye by the reaction of phosphatase in a tissue or cell or on chromosome with a 2'-naphthol AS phosphate, e.g. 3-hydroxy- N-2-biphenyl-2-naphthalenecarboxamide phosphate ester, followed by the reaction between the 2'-naphthol AS phosphate with a diazonium salt; an excitation step for the irradiation of excited light to the azo dye; and a detection step for the detection of fluorescence which is emitted upon irradiation of the excited light. The fluorescence is intense and lasts for a long time.

    This invention relates to a biological indicator derived from a composition comprising: a host organism comprising a spore forming bacteria; a reporter gene for producing an indicator enzyme; a regulatory gene; and a vehicle for inserting the reporter gene and the regulatory gene in the host organism; the host organism bearing a transposable genetic element in its genome for inserting an insertion sequence in the regulatory gene; the insertion sequence comprising a transposase, a pair of terminal inverted repeat sequences, and at least one open reading frame for expressing the transposase. The vehicle may be taken up by the host organism. The insertion sequence may be inserted in the regulatory gene. The host organism may undergo sporulation to form the biological indicator. A process and an apparatus for using the biological indicator are disclosed.

    A rapid method of determining the efficacy of a sterilization cycle, and an indicator adapted to perform such method, comprising subjecting to the sterilization cycle a source of active enzyme having activity which correlates with the viability of a microorganism commonly used to monitor sterilization, and incubating the enzyme source, following the completion of the sterilization cycle, with an effective amount of a substrate system capable of reacting with any residual active enzyme to produce a detectable enzyme-modified product.
