    Abstract

    Early diagnosis of allograft rejection helps to improve the immune‐related management of transplant recipients. The clinically‐used core needle biopsy method is invasive and subject to sampling error. In vivo fluorescence imaging for monitoring immune‐related processes has the advantages of non‐invasiveness, fast feedback and high sensitivity. Herein, we report a responsive second near‐infrared (NIR‐II) fluorescent nanosensor (ErGZ) to detect early allograft rejection. ErGZ allows ratiometric in vivo fluorescence sensing of granzyme B, which is overexpressed in recipients’ T cells during the onset of rejection. The sensor demonstrates efficacious detection of allograft rejection with high sensitivity and specificity, which accomplishes non‐invasive diagnosis of rejection in skin and deep buried islets transplant mice models 2 d and 5 d earlier than biopsy, by in vivo fluorescence imaging and urinary detection, respectively, providing a valuable approach for therapeutical management.

    摘要 早期诊断异体移植排斥反应有助于改善移植受者的免疫相关管理。临床上使用的核心针活检方法具有创伤性,而且容易出现取样误差。用于监测免疫相关过程的体内荧光成像具有无创、反馈快和灵敏度高等优点。在此,我们报告了一种反应灵敏的第二近红外(NIR-II)荧光纳米传感器(ErGZ),用于检测早期异体移植排斥反应。ErGZ 可在体内对颗粒酶 B 进行比率荧光传感,颗粒酶 B 在排斥反应开始时会在受者的 T 细胞中过度表达。该传感器能有效检测同种异体移植排斥反应,灵敏度和特异性都很高,在皮肤和深埋胰岛移植小鼠模型中,通过体内荧光成像和尿液检测,可分别比活检提前 2 天和 5 天对排斥反应进行无创诊断,为治疗管理提供了一种有价值的方法。

    The synthesis of binary fluoride phases, specifically nanofluorides, by coprecipitation from aqueous solutions is considered in terms of phase equilibria. Phases have been precipitated in NaF-RF3 systems (where R stands for a rare-earth element), MF2-YF3 systems (where M = Ca, Sr, Ba), BaF2-ScF3, and CaF2-BaF2 systems. In the MF2-YF3 systems, unordered metastable congruently soluble phases are precipitated instead of ordered fluorite-like phases. In the BaF2-ScF3 system, the congruently soluble compound Ba3Sc2F12 is precipitated. In the NaF-RF3 systems, incongruently soluble metastable phases with a fluorite-like structure (for R = Er, Lu, Y) and the gagarinite structure (NaGdF4) are formed. In the CaF2-BaF2 system, intermediate phases have not been formed.
  • Branched NaYF<sub>4</sub> Nanocrystals with Luminescent Properties

    In this article, branched NaYF4 nanocrystals have been successfully synthesized via a simple hydrothermal method. On the basis of the analysis of HRTEM and TEM images, the growth modes of the branched structure and further branching behavior have been proposed. The up- and down-conversion luminescence of branched NaYF4:Er3+/Yb3+ and NaYF4:Eu3+ have been characterized. Multiarmed NaYF4 phosphors can be introduced into polystyrene to form composite luminescent polymers because of its special geometrical shape. In conclusion, the luminescent branched particles should be of wide potential application as building blocks in the future nanoscience and nanotechnology.

    Complex fluorides ARF4 (A = Li, Na, K, R = rare earthes) are synthesized by the mechanochemical solid reactions of AF and RF3 powders at room temperature using a planetary ball mill. The reactions proceed with the increase of grinding time and have been completed at 4 h. Transmission electron microscope observation shows that the produced ARF4 powders consist of agglomerates of nano-sized primary particles, which exhibit moderate water solubility. When the ground sample is calcined, the particle size is increased and the water solubility is depressed.
