Catalytic Enantioselective Negishi Reactions of Racemic Secondary Benzylic Halides

This report describes the first enantioselective cross-couplings of racemic secondary benzylic halides, specifically, nickel-catalyzed Negishi reactions of bromides and chlorides. The catalyst components are commercially available and air-stable, and the reaction is not highly oxygen- or moisture-sensitive (it can be set up in the air). The method has been applied to the catalytic enantioselective synthesis of intermediates employed by others in the generation of bioactive compounds (e.g., trikentrin A and an androgen receptor agonist).
2,4-Diamino-5-benzylpyrimidines of the formula ##STR1## wherein R.sup.1, R.sup.2, A, Z and n are as hereinafter set forth, are described. The 2,4-diamino-5-benzylpyrimidines of the invention have useful antibacterial activity. More particularly, they inhibit bacterial dihydrofolate reductase and potentiate the antibacterial activity of sulfonamides.

The present invention relates to nitroaryl-substituted phosphoramide prodrug compounds and methods of producing the same for use in targeting and inhibiting undesirable cell growth or proliferation.
N-oxides of benzylpyrimidines of the formula ##STR1## wherein R.sup.1, R.sup.2 and R.sup.3 are as set forth hereinafter, are described. The 2,4-diamino-5-(4-bromo-3,5-dimethoxybenzyl)-pyrimidine is also described. The compounds referred to above are useful as potentiators of the antibacterial activity of sulfonamides.
