Methods for detecting the absence or presence of microorganisms in a sample rely on specific nucleic acid substrates. In particular, the nucleic acid substrates comprise a first stand, a second strand (downstream in the 5' to 3' direction of the first strand) and a complementary strand that is complementary to the first and second strands such that the first and second strands hybridise with the complementary strand to produce a substantially double stranded structure and which allow polymerase and/or ligase activity to be determined and discriminated from one another. Companion kits and blood culture bottles are also described.
