  摘要：

  Enzymatic and chemical oxidation of 4-S-cysteaminylpheno1 (1) and 4-S-cysteaminylcatechol (2) leads to the formation of the hitherto unknown dihydrobenzothiazine-6,7-quinone 4 via intramolecular cyclisation of the o-quinone 6. Oxidation of 4-S-cysteinylcatechol (3) gives the corresponding dihydrobenzothiazinequinone 9, which undergoes rearrangement with partial decarboxylation to give 6,7-dihydroxybenzothiazine derivatives, isolated as 8 and 5 after reduction and acetylation of the mixture. Oxidation of 2 and 3 with periodate or iodate gives mainly the iodinated quinones 7 and 13, respectively.

  摘要：

  A mechanism is proposed for the formation of cyclic 5,6,7,8,9-pentathiabenzocycloheptene-1,2-diol, 4, from the reaction of o-benzoquinone with reduced elemental sulfur, H2Sx. 1,6-Conjugate addition to the quinone is favored over 1,4-conjugate addition. Hydrogen bonding to the quinone oxygen enhances the nucleophilicity of H2Sx by facilitating the removal of the S-H proton. We propose that initially formed 3-polysulfidobenzene-diol intermediates are oxidized to their corresponding quinones and closure of the polysulfur ring subsequently takes place at the C3-C4 bond leading to 4. A possible mechanism for the formation of the pentasulfur linkage in 4 is discussed, which is the key moiety found in a number of natural products.

  An improved synthesis and in vitro activity of cephalosporins with a (4-carboxy-3-hydroxy-5-isothiazolyl)thiomethyl group at the 3-position and its application to the preparation of new derivatives are described. These compounds showed excellent activity against Gram-negative bacteria including β-lactamase producing strains. Among them, 2f was the most interesting because of its broad spectrum of antibacterial activity, including Gram-positive bacteria, and its outstanding inhibitory potency against Pseudomonas aeruginosa.
