Design and synthesis of nonpeptide angiotensin II receptor antagonists featuring acyclic imidazole-mimicking structural units

Extensive molecular modelling studies, including conformational analysis and the comparison of molecular electrostatic potential distributions, were used to evaluate structural parameters of new antagonists containing acyclic replacements of the N = C-N imidazole region. The synthesis and the biological screening of a series of acyl biphenyltetrazole derivatives were planned and realized to gain an insight into the structure-activity relationships of this unusual class of Angiotensin II antagonists. A Facile and General, One-pot Synthesis of 2-Oxoalkane Phosphonates from Diethylphosphonocarboxylic Acid Chlorides and Organometallic Reagents

An efficient and general method for the preparation of 2-oxoalkane phosphonates 2 is described. Diethylphosphono-2-alkanoyl chlorides are used to introduce directly the 2-oxophosphonate 1-substituted synthons on Grignard or cuprate reagents.
A new, practical, synthesis of β-ketophosphonates relying on the conversion of the organolithium reagentfrom a dialkyl methylphosphonate into the correspondingorganocopperreagent, and its reaction with acylchlorides is described. The structure of the intermediate organocopperreagents is discussed.
The silver‐catalyzed oxidative C(sp3)−H/P−H cross‐coupling of 1,3‐dicarbonyl compounds with H‐phosphonates, followed by a chemo‐ and regioselective C(sp3)−C(CO) bond‐cleavage step, provided heavily functionalized β‐ketophosphonates. This novel method based on a readily available reaction system exhibits wide scope, high functional‐group tolerance, and exclusive selectivity.

Treatment of alkynes with diethylphosphite and t-butyl hydroperoxide in the presence of [Cu(MeCN)4]BF4 under microwave irradiation produced the oxyphosphorylation of the triple bond, giving rise to the corresponding β-ketophosphonates in moderate-to-good yields.
