  人5-脂氧合酶（5-LOX）是针对哮喘和炎症的关键酶。Zileuton是唯一一种针对5-LOX的药物，由于几个问题而退出市场。在本研究中，报道了我们先前报道的2-氨基-4-芳基噻唑（1）对噻唑（2）和硫脲（3）支架进行合理设计的偶联物的性能。它们是合成的（总共31个衍生物），在体外进行了表征并针对5-LOX酶进行了测试，并确定了活性最高的酶的作用方式。化合物2m的IC 500.9±0.1μM的化合物通过竞争性（非氧化还原）机理起作用，与Zileuton不同，它没有自由基清除性能。计算研究与支持其作用机理的实验数据非常吻合。来自硫脲系列（3）的另一个铅分子3f对5-LOX的IC 50为1.4±0.1μM，其作用方式为氧化还原类型（非竞争性）。有希望地注意到，铅抑制剂2m和3f表现出的活性均优于商品药物Zileuton（IC 50  = 1.5  ±  0.3μM）。这些抑制剂可以进一步发展为抗炎药。

  AbstractThe reaction between secondary amines, benzoyl isothiocyanate, and dialkyl acetylenedicarboxylates (=dialkyl but‐2‐ynedioates) in the presence of silica gel (SiO2) led to alkyl 2‐(dialkylamino)‐4‐phenylthiazole‐5‐carboxylates in fairly high yields. The structures of the products were confirmed by their IR, 1H‐ and 13C‐NMR, and mass spectra, and by a single‐crystal X‐ray structure determination.

  The complexes of 8 N,N-disubstituted,N'-benzoyl thioureas with copper, nickel and cobalt have been studied. NMR spectra and magnetic measurements have shown square planar NiL2 and fac CoL3 compounds. The electrochemical parameters obtained from cyclic voltammetry for the irreversible process copper(II)/copper(I) redox systems are slightly influenced by the different substituents.
