Application of [3 + 2]-Cycloaddition in the Synthesis of Valdecoxib
摘要:
A large scale synthesis of valdecoxib 1 is described. Our work features potential application of [3+2]-dipolar cycloaddition involving enamine and in situ-generated nitrile oxide derivatives.
A general access to isoxazoles with outstanding functional group compatibility from the readily available ynones using trimethylsilyl azide as an amino surrogate under exceptionally simple conditions is described.

An efficient method for the synthesis of 3,5-disubstituted Isoxazoles is described. The reactions of 1,3-dialkynes with hydroxylamine proceeded smoothly in DMSO under mild reaction conditions to produce 3,5-disubstituted isoxazoles In satisfactory to excellent yields.
