Crystallographic analysis of several non-limiting chromophores reveals, for instance, large ring-ring dihedral twist angles and a highly charge-separated zwitterionic structure in the ground state, in both solution phase and solid-state.

    The presently disclosed subject matter provides dyes having an improved photostability, biosensors comprising such dyes, and methods of use thereof, including methods for detecting target molecules in a sample under test and for live-cell imaging. The dyes can include a binding member, including a biomolecule or fragments thereof, which can interact with target molecules of interest and can be specific to a given conformational state or covalent modification, e.g., phosphorylation, of the target molecule. The presently disclosed dyes can be used for detecting changes in the binding, conformational change, or posttranslational modification of the target molecule.

    A method of imaging using selective fluorescent emitters and compositions for imaging are described. The method can include contacting a specimen with a composition comprising at least one selective fluorescent emitter of Formulas I through IX and irradiating the specimen with an excitation wavelength. The fluorescence emitted by the selective fluorescent emitter can be detected to generate an image. Formulas I through IX are: wherein: R 1 =OR 4 or NR 3 R 4 ; R 2 =OR 4 , NR 3 R 4 , Cl, F or H; R 3 =H, CH 3 , C 2 H 5 , C 3 H 7 or C 4 H 9 ; R 4 =H, CH 3 , C 2 H 5 , C 3 H 7 or C 4 H 9 ; R 5 =H or CH 2 (CH 2 ) n NR 3 R 4 ; R 6 =H or CH 2 (CH 2 ) n NR 3 R 4 ; R 7 =OR 4 , NR 3 R 4 , Cl, F or H; R 8 =N-methylpyridinium or N-methyl-1,2,3,6-tetrahydropyridine; and n=1, 2 or 3.
