硅烷试剂

硅烷试剂是一类用于变更或保护有机物中活性基团的有机硅单体或小分子化合物,主要用于分析和有机合成领域,如医药合成等。反应后,硅烷基会脱落恢复原状。目前已知有500余种不同类型的硅烷试剂,主要包括以下几类:

  1. 三甲基甲硅烷基单官能SBA,例如三甲基氯硅烷、六甲基二硅氮烷。
  2. 位阻型单官能SBA,如叔丁基二甲基氯硅烷、三异丙基氯硅烷。
  3. 位阻型双官能SBA,例如二甲基二乙酰氧基硅烷、二叔丁基二氯硅烷。
  4. 其他保护剂,比如三甲基羟乙基硅烷和甲基二苯基羟乙基硅烷。

这些试剂的应用范围广泛,除了用于分析领域外,还可以用作改性药物,如使其成油溶性或丧失苦味等。硅烷化作用是指将硅烷基引入到分子中,通常是取代活性氢(如:羟基-OH、羧基-COOH、氨基-NH2、巯基-SH、磷酸盐)。

通过硅烷化反应后,化合物的极性会降低,减少了氢键束缚,使得形成的硅烷化衍生物更容易挥发;同时由于含活性氢的反应位点数目减少,化合物的稳定性也得以加强。硅烷化使被测物能力增强,热稳定性提高。许多被认为是不挥发或在200-300℃下不稳定羟基或氨基化合物,在进行硅烷化处理后成功进行了色谱分析。

由于硅烷化试剂对活泼氢非常敏感,因此它们也容易受到潮气的影响而分解失效。为了确保其有效性,通常将硅烷化试剂密封保存在氮气环境中,并且必须提前除去氮气中的水分。使用时,在再次密封前应立即充入氮气以避免空气接触。具体操作方法如下:连接特氟龙管的一端至氮气钢瓶,另一端连巴氏吸管;将吸管伸入试剂瓶内约2~3cm处,缓缓开启氮气开关直到充满空白空间后封紧瓶子并关闭氮气。

三甲基硅咪唑是高效的羟基和羧基的硅烷化试剂,能快速且平滑地与这些官能团反应。它不与胺或酰胺发生反应,因此适合用于制备含有羟基和氨基化合物的多重衍生物。即使在少量水存在的情况下也能进行糖类的硅烷化,特别适用于将糖作为糖浆剂进行分析。这种试剂能够衍生大多数未被阻碍及严重阻碍的甾类羟基。
