A method for the determination of cells in a sample is disclosed. The sample is typically urine and the method is useful in removing interfacing reductants from the sample. The method comprises the steps of:
1) separating the cells from the sample,
2) washing the separated cells with:
a) an iron(III) chelate solution and
b) a non-ionic surfactant solution and
3) contacting the washed cells with a redox reagent so as to produce a detectable change due to the presence of the cells.

Reagents and methods for the determination of analytes are disclosed, using water soluble cobalt(III) complexes and metallizable dyes to form cobalt(III) complexes of the metallizable dyes. Analytes which can be determined include enzymes, cells and biological reductants, such as NADH, NADPH and ascorbate. Amplification is provided since one equivalent of the analyte provides more than one equivalent of the detectable species.

Cobalt(III) reagents in combination with water soluble polymers

Water soluble polymers are useful in combination with cobalt(III) redox reagents. When incorporated with the polymers, the cobalt(III) reagents exhibit improved sensitivity compared to fresh solution of the reagents. In preferred embodiments, the cobalt(III) reagents are coated on a support using the water soluble polymer as a binder. Small areas of the coated support are then cut out and placed in a buffer solution to provide a working reagent solution. The solution is particularly useful in the testing of urine for significant bacteriuria.
To provide an immunochromatographic kit which realizes highly sensitive measurement for which an exclusive analyzer is not required.
The immunochromatographic kit includes an inspection strip (1) which includes an insoluble carrier (2) spreading the specimen liquid, a label-holding pad (3) including a label substance modified with a first substance bondable to a test substance, a liquid-sending pad (4) sending a first amplification liquid (41) to the insoluble carrier (2), and an absorption pad (5) disposed in contact with the other end of the insoluble carrier (2) and sequentially has an inspection region (L1) including a second substance being bonded to the test substance, a confirmation region (L2) including a substance bondable to the first substance, and an amplification index region (L3) including a substance being reacted with the first amplification liquid between the label-holding pad (3) and the absorption pad (5) of the insoluble carrier (2), a first pot (40) being disposed below the liquid-sending pad (4) and enclosing the first amplification liquid (41), and a second pot (50) being disposed above the absorption pad (5) and enclosing a second amplification liquid (51) in a housing case (9).
