The reaction of N-tosyliminoisoquinolinium ylides with trimethylsilylketene as a C2 unit introducing reagent, giving unexpected [3+2] cycloadducts, pyrazolo[5,1-a]isoquinolines, is described.

Piperazinylmethyl substituted pyrazolo[1,5-a]pyridines and related heterocycles were synthesized and found to recognize selectively the dopamine D4 receptor. For the most potent derivative 10d a Ki value of 2.0 nM was observed. SAR studies including the comparison of molecular isopotential surfaces were performed. (C) 1998 Elsevier Science Ltd.
