Disclosed are compounds of Formula (I), and pharmaceutically acceptable salts thereof. The compounds of formula (I) inhibit tyrosine kinase activity of JAK3, thereby making them useful for the treatment of inflammatory and autoimmune diseases.

The synthesis and selected transformation of some cyclopentanoid β-ketoximesulfone derivatives are reported. Utilization of the sulfone group to stabilize adjacent negative charge facilitates smooth alkylation. Subsequent 1,4-elimination of the sulfonyl moiety generates a vinyl nitroso species that is readily intercepted by lithium acetylides.

We have developed a new iron-catalyzed ring opening alkenylation of cycloketoximes with β-nitrostyrenes for the facile synthesis of flexible distal cyanoalkyl alkenes.
