  本发明提出一种帕罗韦德中间体(1S,3aS,4aR)‑2‑[(2S)‑3,3‑二甲基‑1‑氧亚基‑2‑[(三氟乙酰基)氨基]丁基]‑4,4‑二甲基‑2,3,3a,4a‑四氢‑1H‑环丙并[1,2‑c]吡咯‑1‑甲酸(I)的合成方法。该方法以(1S,3aS,4aR)‑4,4‑二甲基‑2,3,3a,4a‑四氢‑1H‑环丙并[1,2‑c]吡咯‑1‑甲酸甲酯(III)和L‑叔亮氨酸(V)为原料，先将原料(III)水解成(1S,3aS,4aR)‑4,4‑二甲基‑2,3,3a,4a‑四氢‑1H‑环丙并[1,2‑c]吡咯‑1‑甲酸(II)，将L‑叔亮氨酸(V)用三氟乙酰基保护得到N‑三氟乙酰基‑L‑叔亮氨酸(IV)；最后中间体(II)与中间体(IV)缩合，即可得到化合物(I)。本发明采用汇聚式合成路线，只需要三步反应就可以得到化合物(I)，缩短了合成路线，为化合物(I)的制备提供了新的方法。

  Alkyl fluoroformates RHOC(O)F are a new family of excellent alkylating agents. Perfluoroacyl fluorides, selected perfluoroketones and fluoroolefins can be alkylated to hydrofluoroethers RF–O–RH and hydrofluoroalkanes RF–RH, respectively. Potassium fluoride and cesium fluorides catalysts in glymes at 50–100 °C are the preferred experimental conditions.
