Lipase-catalyzed regioselective preparation of fatty acid esters of hydrocortisone

A series of fatty acid derivatives of hydrocortisone has been prepared by an enzymatic methodology. Nine 21-monoacyl products and one 3,11,17-triacetyl derivative, nine of them novel compounds, were obtained in a highly regioselective way through lipase-catalyzed esterification, transesterification and alcoholysis reactions. The influence of various reaction parameters Such as acylating agent: substrate ratio, enzyme: substrate ratio. solvent, temperature and nature of acylating agent and alcohol was evaluated. Among the tested lipases. Candida antarctica lipase appeared to be the most appropriate and showed a high efficient behavior especially in a one-pot transesterification. The advantages presented by this methodology, such as mild reaction conditions and low environmental impact, make the biocatalysis a convenient way to prepare acyl derivatives of hydrocortisone. A steroid having an anti-inflammatory activity can be administered as a fat emulsion thereof. The emulsion may comprise an effective quantity of the steroid having an anti-inflammatory activity, 5 to 50% (W/V) of soybean oil, a phospholipid in a weight ratio of 1-50 to 100 of said soybean oil, and a proper quantity of water. This fat emulsion exhibits a long-lasting activity in the region of inflammation and is especially useful in the therapeutic or prophylactic treatment of rheumatism, immunological hemolytic anemia, idopathic thrombocytopenic purpura, and Paget's disease, or in conjunction with kidney transplantation.

A kit of a medicine-containing fat emulsion of the type prepared immediately before its use, which comprises a fat emulsion and either (a) a medicinal composition comprising a medicine and at least one solvent selected from among liquid polyalkylene glycols, liquid alkylethanolamines, liquid polyhydric alcohols and water, or (b) a medicinal composition comprising a medicine and an excipient such as a sugar and/or an amino acid, and a process for preparing a medicine-containing fat emulsion therefrom immediately before its use.
