Bis(dialkylaminethiocarbonyl)disulfides as Potent and Selective Monoglyceride Lipase Inhibitors

Monoglyceride lipase (MGL) inhibition may offer an approach in treating diseases in which higher 2-arachidonoyglycerol activity would be beneficial. We report here the synthesis and pharmacological evaluation of bis(dialkylaminethiocarbonyl)disulfide derivatives as irreversible MGL inhibitors. Inhibition occurs through interactions with MGL C208 and C242 residues, and these derivatives exhibit high inhibition selectivity over fatty acid amide hydrolase, another endocannabinoid-hydrolyzing enzyme.
Additionally, 3-methyl-1H-indol-1-yl dimethylcarbamodithioate (3o) effectively ameliorated histopathological changes of lung tissues and attenuated lipopolysaccharides (LPS)-induced acutelunginjury (ALI) in vivo. These data suggest that the new indole-dithiocarbamate derivatives could be particularly useful for further pharmaceutical development for the treatment of ALI.

The invention relates to telechelic polymers based on radically polymerized, ethylenically unsaturated monomers which are obtained by polymerization by means of thiuram disulfides and dithiocarbamates, which have one or several additional functional reactive groups in addition to the carbamate group. The invention furthermore relates to the manufacture of the said type of telechelic polymers and to the use thereof, for example, as reactive precursors in higher-polymer linear or cross-linked products, especially as components of aqueous solutions, emulsions, dispersions, and suspensions for the preparation of coatings.
Die Erfindung betrifft telechele Polymere auf Basis von radikalisch polymerisierten äthylenisch ungesättigten Monomeren, die durch Polymerisation mittels Thiuramdisulfiden und Dithiocarbamaten erhalten werden, welche neben der Carbamat-Gruppierung eine oder mehrere weitere funktionelle reaktive Gruppe(n) aufweisen. -Die Erfindung betrifft ferner die Herstellung derartiger telecheler Polymerer sowie ihre Verwendung, beispielsweise als reaktive Vorstufe von höherpolymeren linearen oder vernetzten Produkten, insbesondere als Bestandteil von wäßrigen Lösungen, Emulsionen, Dispersionen und Suspensionen für die Herstellung von Beschichtungen.
