A fluorescent rosamine dye having specificity for neural stem cells, specifically, proliferative neural stem cells, and represented by Structural Formula (I): wherein X is an anion, is described herein. Synthesis of the fluorescent rosamine dye and use of the fluorescent rosamine dye to detect symmetric and asymmetric division of neural stem cells are also described.

We report the first solidphasesynthesis of a xanthone library CX and its application to embryonic stem cell probe development. The CX library was further derivatised with an activated ester resin to provide an acetylated CX (CXAC) library. Screening of these libraries led to the discovery of a novel fluorescent mESC probe, CDb8.

Xanthones and acridones were synthesized from 3,4-difluoronitrobenzene and 2-fluorobenzaldehydes in two or three steps. The key step was nucleophilic aroylation catalyzed by imidazolidenyl carbene. The nucleophilic aroylation of 3,4-difluoronitrobenzene afforded 2,2'-difluoro-4-nitrobenzophenones. The cyclization of the difluorobenzophenones with O-nucleophile and N-nucleophile yielded 3-nitroxanthones
