    New palladium complexes with bis(8-thiotheophylline)alkane derivatives

    Aiming at finding new models that allow us to obtain more information about the interaction between metals and purine-biomolecules, the new derivatives bis(S-8-thiotheophylline)methane (MBTTH2) (1), 1,2-bis(S-8-thiotheophylline)ethane (EBTTH2) (2), 1,3-bis(S-8-thiotheophylline)propane (PBTTH2) (3) and, 1,10-bis(S-8-thiotheophylline)decane (DBTTH2) (4) were synthesised. The evaluation of the coordinative properties of these new ligands against cis-[PdCl2(PPh3)(2)], leaded to the synthesis of the complexes trans-[PdCl(PPh3)(2)LH] (L = EBTT2- (5), PBTT2- (6)) and trans-[(PdCl(PPh3)(2))(2)L] (L = MBTT2- (7), EBTT2- (8), PBTT2- (9), DBTT2- (10)). All of the ligands and complexes were characterised by elemental analysis, IR and multinuclear (H-1, C-13{H-1}, P-31{H-1} NMR) spectroscopy. The crystal structure of the ligand 1 is also presented, confirming the proposed composition for the new purine derivatives. (C) 2003 Elsevier B.V. All rights reserved.

    The new water-soluble ruthenium(II) mononuclear complexes [RuCp(X)(PTA)(L)] (X = 8-thio-theophyllinate (TTH-), L = PTA (1), L = PPh3 (7)); (X = 8-methylthio-theophyllinate (8-MTT-), L = PTA (2), L = PPh3 (8)), (X = 8-benzylthiotheophyllinate (8-BzTT(-)), L = PTA (3), L = PPh3. (9)) and binuclear complexes [RuCp(PTA)(L)}(2)-mu-(Y-kappa N7,N'7)] (Y = bis(S-8-thiotheophyllinate)methane (MBTT2-), L = PTA (4), L = PPh3 (10)), (Y = 1,2-bis(S-8-thiotheophyllinate)ethane (EBTT2-), L= PTA (5), L=PPh3 (11)), (Y=1,3-bis(S-8-thiotheophyllinate)propane (PBTT2-); L = PTA (6), L=PPh3 (12)) have been synthesized and characterized by NMR, IR spectroscopy and elemental analysis. The single crystal X-ray structure of [RuCp(8-MTT-kappa S)(PTA)(2)] (2) was also obtained. The antiproliferative activity of the complexes on cisplatin-sensitive T2 and cisplatin-resistant SKOV3 cell lines has been evaluated.

    Thiopurines bis(S-8-thiotheophylline)methane (MBTTH2), 1,2-bis(S-8-thiotheophylline)ethane (EBTTH2), and 1,3-bis(S-8-thiotheophylline)propane (PBTTH2) were reacted with [RuClCp(mPTA)(2)](CF3SO3)(2) in water to afford the bis-ruthenium complexes [RuCp(mPTA)(2)}(2)-mu-(L-N7,N7)](CF3SO3)(4) (1: L = MBTT; 2: L = EBTT; 3: L = PBTT), which have been characterized by elemental analysis, IR, and multinuclear NMR (H-1, C-13H-1}, P-31H-1} and F-19H-1}) spectroscopy). Diffusion experiments for 1 were carried out. Proposed structures for the three complexes were also supported by theoretical calculations. Cyclic voltammetry showed that these complexes are characterized by two one-electron irreversible oxidative response (Ru-II - Ru-II/Ru-III -Ru-II; Ru-III - Ru-II/Ru-III - Ru-III). Complexes showed poor antiproliferative activity against cisplatin-sensitive T2 human cell line and the cisplatin-resistant SKOV3 cell line.
