The present invention provides a method for extracting a nucleic acid from a sample and depleting inhibitors contained in the sample, comprising:
a) preparing an extraction mixture by contacting the sample with
i) at least one cationic detergent capable of forming a precipitate with inhibitors contained in the sample,
ii) at least one salt, wherein the salt is contained in the extraction mixture in a concentration to prevent precipitation of the nucleic acid in the presence of the cationic detergent, and
iii) at least one water-insoluble non-ionic polymeric adsorbent;
b) incubating the extraction mixture and forming a precipitate which comprises the cationic detergent and inhibitors contained in the sample and separating the formed precipitate from the supernatant, thereby providing an inhibitor depleted supernatant which comprises the extracted nucleic acid.
Also provided is a kit for performing said method.
