  New pyrazolo-pyrrolo-pyrimidine-dione (PPPD) compounds of formula (I), ##STR1## where R.sub.1, R.sub.2, R.sub.3, R.sub.4 and m are as defined in the specification, e.g., 4-(benzoylmethyl)-6-cyclohexyl-6,7-dihydro-2-phenyl-4H-pyrazolo[1,5-a]pyrr olo[3,4-d]pyrimidine-5,8-dione, have been found to be potentially useful to treat warm-blooded animal patients suffering from the symptoms of atherosclerosis and cholesterol buildup to relieve the same and for favorably altering the high density lipoprotein (HDL) to low density lipoprotein (LDL) ratio in blood samples of such patients.

  An amino blocking reagent, 1-isopropyl-3-ethoxy-4-nitro-2-oxo-3-pyrroline has been obtained. The new reagent is water soluble. It has yielded an N-protected glycine active ester with solubiity appropriate for reaction with both peptides and proteins. N-hydroxysuccinimide esters of N-i-NOPY- and N-c-NOPY-glycines were prepared. The N-hydroxysuccinimide active esters were produced by treatment with dicyclohexylcarbodimide (DCC) and N-hydroxysuccinimide in tetrahydrofuran (THF). The new reagents for glycine attachment are stable crystalline compounds. These active esters were used to attach glycine residues to a dipeptide and a protein by the salt-coupling procedure.

  2-Cyclohexyl-1,3,4,5-tetrahydro-10H-pyrrolo[3,4-b][1,5]benzodiazepine- 1,4-dione was synthesized by heating a solution of ethyl 1-cyclohexyl-2,3-dioxopyrrolidine-4-carboxylate and o-phenylenediamine in acetic acid. Alkylation and acylation of the synthesized compound were examined. 4-Aryl-2-cyclohexyl-1,3,4,10-tetrahydropyrrolo[4,3-c][1,5]benzothiazepin-1-ones were synthesized by Michael addition of 2-aminothiophenol to 4-arylidene-1-cyclohexyl-2,3-dioxopyrrolidines and concomitant dehydrative cyclization.
