The partial reduction of heterocycles: an alternative to the Birch reduction

The partial reduction of a series of heterocycles in THF is described. By adding amine 3 as a proton source and naphthalene as an electron carrier, we were able to produce reductively alkylated pyrrolines and dihydrofurans in moderate to good yields. This reaction does not require liquid ammonia as a solvent, which may have interesting ramifications for large-scale synthesis. Moreover, we were also able to quench the reduction reactions with an acid chloride so performing a reductive acylation reaction. (C) 2000 Elsevier Science Ltd. All rights reserved.

The synthesis and directed dihydroxylation of a range of cyclic alkenes was investigated. Both homoallylic alcohols and homoallylic trihaloacetamides were found to be efficient directing groups, giving rise to good to excellent levels of remote asymmetric induction with OsO4–TMEDA. Interestingly, in all cases examined, trifluoroacetamides were found to be superior to trichloroacetamides as directing groups and an argument is presented which rationalises this observation.

The synthesis of 2-furoic acid derivatives containing both 3-methoxy and 3-TMS groups is described. Reductive alkylation proceeded well to give us access to a series of highly functionalised dihydrofurans with potential for further elaboration. Of the two groups tested at C-3, the TMS derivative was found to be the more useful and gave rise to high levels of stereoselectivity when attached to a furan
