    The present disclosure provides a method for analyzing a functional subunit pair gene of a T cell receptor (TCR) or a B cell receptor (BCR). The present disclosure provides a method comprising: (1) a step for providing a nucleic acid sample containing a nucleic acid of a TCR or BCR from a cell expressing an activated TCR or BCR; (2) a step for determining the nucleic acid sequence of the functional pair gene of the TCR or the BCR; and (3) a step for calculating the frequency of appearance of each gene and combinations thereof on the basis of the determined nucleic sequences, and identifying a functional subunit pair gene of the TCR or the BCR.

  • Renilla based biosensors for monitoring biomolecule localization and trafficking in cells

    Bioluminescence resonance energy transfer (BRET) biosensors for assessing the intracellular localization, internalization and trafficking into cellular compartments of proteins such as receptors, and other biomolecules such as second messengers, are disclosed. These biosensors, which are dependent on the concentration/density of the BRET donor and acceptor in cellular compartments rather that specific protein-protein interactions, use a Renilla GFP/Luc BRET pair, which allows the robust and reproducible monitoring of protein trafficking/localization, with a sensitivity compatible with high-throughput screening (HTS). The use of these biosensors for various applications, including assessing/monitoring protein endocytosis, recycling and intracellular trafficking, receptor maturation/rescue by pharmacological chaperones, various endocytosis/exocytosis processes, activation/inhibition, as well as biomolecule concentration/density in different cellular compartments, is also disclosed.

    The invention relates to an assay for the identification of a compound having immunosuppressant activity, wherein a candidate compound is analyzed whether it blocks the Ca 2+ flux in coronin 1 expressing cells and/or in coronin 1 negative cells. A candidate compound is identified as having immunosuppressant activity if it blocks the Ca 2+ flux specifically in coronin 1 expressing cells. Further described are upstream assays wherein the impact of a candidate compound on coronin 1 trimerization is measured, and downstream assays wherein the impact of a candidate compound on diacyl glycerol (DAG) generation, phosphatidylinositol-4,5-biphosphate (PIP 2 ) levels and/or inositol-1,4,5-triphosphate (InsP 3 ) generation or on nuclear factor of activated T cells (NFAT) nuclear localization is determined.
