    High-intensity ultrasound (HIU) and microwave (MW) irradiation, having emerged as effective promoters of organic reactions, were exploited for the synthesis of bile acids derivatives. Esterification, amidation, hydrolysis, oxidation, and reduction were investigated. Compared to conventional methods, both techniques proved much more efficient, increasing product yields and dramatically cutting down reaction times. Scaled-up studies are now under way. (C) 2004 Elsevier Inc. All rights reserved.

    Process for preparing glycocholic acid comprising the following steps: a) making a mixed anhydride of cholic acid through the suspension of cholic acid in acetone or in aqueous acetone in presence of a base, and subsequent reaction with an alkyl chloroformate; b) aminolysis of the so obtained mixed anhydride of cholic acid with an aqueous solution of a glycine ester, resulting in an ester of glycocholic acid; c) hydrolysis of the ester of glycocholic acid thus obtained through the treatment with aqueous soda or aqueous potassium and precipitation of glycocholic acid through acidification of the corresponding sodium salt.
    制备胆酸的过程包括以下步骤:a) 通过在丙酮或水合丙酮中悬浮胆酸,并在碱的存在下制备胆酸的混合酐,然后与烷基氯甲酸酯反应;b) 将得到的胆酸混合酐与甘氨酸酯的水溶液进行氨解反应,得到胆酸的酯;c) 通过用水合钠或水合钾处理得到的胆酸酯,使其水解并通过酸化相应的钠盐沉淀出胆酸。

    本发明涉及一种甘氨胆酸的制备方法,以N,N‑羰基二咪唑(CDI)为缩合剂和纳米氧化钙为缚酸剂,对原料胆酸进行活化处理,所得活性中间体具有良好的稳定性,与甘氨酸乙酯盐酸盐进行缩合反应制备甘氨胆酸乙酯,再进行水解反应得到目标产物甘氨胆酸,具体合成路线如下。采用本发明的方法制备甘氨胆酸,不易发生副反应,制备方法简单,杂质含量低,提高了反应效率,收率和纯度较高,收率达到90%以上,纯度达到99%以上,后处理简单,不需要柱层析纯化,生产成本低,适合工业化大规模生产。
