  Novel [(arylcarbonyl)oxy]propanolamines were synthesized and investigated as potential ultrashort-acting beta-adrenergic receptor blockers. Many of these analogues exhibited good potency and short duration. The N-ureidoalkyl analogue 85 (ACC-9089) has a potency equal to propranolol and a duration of action of about 21 min in the dog. It has been selected as a candidate for further clinical study. Structure-activity relationships and structure-duration relationships for these new beta-blockers are also discussed.

  The invention provides a method for the treatment or prophylaxis of cardiac disorders in a mammal, comprising administering to such mammal a short-acting β-blocking compound of the formula wherein X is. is lower alkyl, lower alkenyl or aralkyl; R, is lower alkyl; and Ar is unsubstituted aromatic or aromatic substituted with lower alkyl, lower alkenyl, lower alkynyl, lower alkoxy, halogen, acetamido, amino, nitro, alkylamino, hydroxy, hydroxyalkyl, cyano, or a group of the formula wherein R2 is lower alkyl, aryl, or aralkyl and n is an integer from 0 to about 10; or a pharmaceutically acceptable salt thereof. Furthermore the invention provides compounds of the general formula wherein X is Ris lower alkyl, or aralkyl; R, is lower alkyl; and Ar is unsubstituted aromatic or aromatic substituted with lower alkyl, lower alkenyl, lower alkynyl, lower alkoxy, halogen, acetamido, amino, nitro, alkylamino, hydroxy, hydroxyalkyl, cyano, or a group of the formula wherein R2 is lower alkyl, aryl, or aralkyl and n is an integer from 0 to about 10; or a pharmaceutically acceptable salt thereof.
