  Certain embodiments of the present invention provide compounds that comprise at least one guanidine moiety and have a binding affinity for the voltage sensing domain (VSD) of a voltage-gated ion channel, such as the Hv1 voltage-gated proton channel. In some embodiments, the compounds of the present invention that have as VSD binding affinity are further characterized by having a property of inhibiting ion transport by the voltage-gated ion channel comprising the VSD, such as inhibiting proton transport by Hv1. In some embodiments, the compounds according to the invention comprise at least one guanidine moiety in a five-membered aromatic ring.
