    Oxidation reactions using air as oxidant thanks to silica nanoreactors containing GOx/peroxidases bienzymatic systems

    Anew kind of enzymes encapsulation method is presented It combines the sol-gel method with a templating process using Mayers of phospholipids to provide an organized network of phospholipids inside the silica and in the same time protect the embedded enzymes as if they were entrapped in a biological membrane supported on silica This brings a porosity control to the classical sol-gel encapsulation and therefore increases the accessibility of the substrates to a maximum of enzymes A first family of hybrid phospholipids-silica materials named Sponge mesoporous silica (SMS) has been synthesized which features a sponge-like structure The formation of a negative curvature in the rigid phospholipid Mayers was induced by adding ethanol (>50%) to the aqueous medium which provokes the separation of the head groups and the undulations of the lipid Mayers Dodecylamine added to stabilize this structure acts as a catalyst for silica condensation Lactose was added to avoid direct interaction of the enzymes with the silica at the interface with the lipids bilayer Lipase was encapsulated in SMS and shows higher activity for ester hydrolysis compared to commercial immobilized-lipase An experiment design plan was carried out to optimize SMS synthesis using hemoglobin (Hb) as biocatalyst for oxidation reactions using H2O2 As a result a new structure was identified formed by the aggregation of nanoporous silica capsules named NPS with a higher activity compared to SMS biocatalysts These new biological nanoreactors were used to encapsulate simulteanously Hb and glucose oxidase to produce in situ H2O2 from air and perform oxidation reactions This system was used for the elimination of the carcinogenic polycyclic aromatic hydrocarbon pollutants from water (C) 2010 Elsevier B V All rights reserved

    Provided herein are methods for enhancing enzymatic activity using certain polymers that may be optionally attached to an enzyme. The polymers may be thermally-responsive polymers, including poly N-isopropylacrylamide or poly N-isopropylmethacrylamide. The polymer may also be a copolymer with at least two different monomer residues. The monomer residues may have a structure of formula (I): wherein R 1 , R A and R B are as described herein. Examples of such monomer residues may include N-isopropylacrylamide (NIPAm) or N-isopropylmethacrylamide (NIPMa). The polymer may include additional monomer residues, such as aminooxy-bearing methacrylamide monomer residues that can be modified to vary the lower critical solution temperature (LCST) of the polymer.

    New ceramide trafficking inhibitors, (1R,3R)-N-(3-hydroxy-1-hydroxymethyl-3-phenylpropyl)dodecanamide (HPA-12) and a series of its analogues, were synthesized in diastereomerically and enantiomerically pure forms, and the structure-activity relationship was investigated. These analogues were stereoselectively synthesized via catalytic enantioselective Mannich-type reactions using a Cu(II)-chiral diamine 4 complex. Analysis of HPA-12 analogues having various lengths of the amide side chain showed that the optimal chain length for the inhibition of sphingomyelin biosynthesis is 13 with an IC50 of similar to50 nM. Masking of the hydroxy group at the 2'- or 3-position of HPA-12 was carried out by methylation, and it was revealed that these hydroxy groups were essential for the activity. Installation of another hydroxy group onto HPA-12 at the same position as that in the natural ceramide was also conducted, but no enhancement of the activity was observed.
