谷氨酸类衍生物

学名为α-氨基戊二酸,最初从谷物中提取而得名。它分布广泛,几乎存在于所有蛋白质组分中。通过蛋白质水解即可获得谷氨酸。这种物质为无色板块状结晶,等电点为3.3,易溶于水而不溶于苯、醚等有机溶剂;其水溶液呈酸性,因此被归类为酸性氨基酸。

在生理上,谷氨酸能够与甘氨酸和半胱氨酸结合形成重要的生物分子——谷胱甘肽。此外,谷氨酸本身具有鲜味特性,其钠盐就是常见的味精(即味精)。值得注意的是,在高温下使用味精时应避免强热分解。日本化学家池田首次发现了味精的这一特性:在一个偶然的机会中,他喝海带汤感到有鲜味,并通过研究从100公斤海带中提取了2克谷氨酸钠。之后,日本大规模生产并将其命名为“味之素”。

中国在20世纪30年代由上海天厨味精厂率先开始生产味精,远销全球而闻名于世。通常,人们会使用天然麦胶蛋白或酪蛋白进行水解,在溶液pH值调至3.3时得到谷氨酸沉淀,再通过添加适量氢氧化钠制成味精。尽管人工合成法成本较高,但目前仍主要采用蛋白质水解法制备。20世纪60年代以后,日本开始利用微生物发酵生产谷氨酸,即短小杆菌属、小球菌属和芽孢杆菌属的细菌在糖类化合物(主要是淀粉或葡萄糖)中分泌大量谷氨酸,用于制造味精。

此外,谷氨酸还具有与血液中的氨结合形成无毒物的功能,并被用作治疗肝昏迷及恢复期肝功能不全等病症。
