木脂素

木脂素是一类由2分子或3分子苯丙基以不同形式聚合而成的天然有机化合物,广泛分布于被子植物和裸子植物中。它们可以分为两大类:一类是由两分子苯丙基衍生物通过其侧链β-位聚合形成的,称为木脂素;另一类则由一分子苯丙基的侧链与另一分子的苯环连接或以氧原子连接而形成的新木脂素。此外,还包括由三分子苯丙基聚合而成的倍半木脂素(sesquilignan),以及由黄酮或查尔酮与苯丙烯衍生物缩合形成的黄酮木脂素(flavonolignan)和查尔酮木脂素(xanthonolignan)。木脂素在两分子苯丙基β-碳缩合后,侧链γ-碳上的含氧基团还能相互脱水缩合形成取代的四氢呋喃、半缩醛、内酯四氢萘或环辛烯等结构。因此,木脂素还可进一步分为单环氧木脂素(monoe-poxy lignan)、木脂内酯(lignanolide)和环木脂素(cycloli-gnan)等多种类型。

目前从植物界已分离出200多个木脂素及其衍生物,其中包括100多种新木脂素。如五味子、刺五加、了哥王、络石、牛蒡子、远志、连翘、红花和细辛等中药中都含有木脂素。

在植物体内,木脂素通常以游离态或甙的形式存在,大多数具有光学活性。游离的木脂素多为无色结晶,某些新木脂素呈油状物;它们可溶于有机溶剂,难溶或不溶于水。甙类化合物在醇溶液中溶解度较大,并且可以被酸水解。提取木脂素时通常先用甲醇或乙醇提出总木脂素,再通过色谱法或分步结晶等方法进行分离。

多数木脂素类成分遇浓硫酸能显色。含有酚羟基的分子可与某些试剂反应呈现不同颜色;例如,鬼臼脂素在三氯化铁溶液中呈棕黄色,α-足叶草脂素则显紫色。此外,一些木脂素还能与五氯化锑的四氯化碳溶液发生反应而显色。

由于部分环状结构可能具有立体异构体,某些中药中存在的木脂素成分性质不稳定,在酸性或碱性的条件下容易发生异构化,转化为立体异构体。例如鬼臼脂素在碱性溶液中易转变为苦鬼臼脂素(Picrop-odophyllin),而受三氯化磷影响时C1-OH会转位生成表鬼臼脂素(Epi-podophyllotoxin);这两种化合物均无抗癌活性。

研究表明,多种木脂素具有抗癌、抗真菌、杀虫、升白细胞、降低谷丙转氨酶、保肝、止咳和泻下等多种生物活性。其中,鬼臼木脂素的衍生物作为抗癌药物被广泛研究和应用;五味子中的木脂素则有降谷丙转氨酶的作用,并用于治疗肝炎。
