[Class 2 (iii)] Amino-acid amidines, including cyclic amidines such as imidozolines and tetrahydropyrimidines, are made by causing a reactive ester of a hydroxyalkylamidine to react with ammonia or a primary or secondary amine. The reactive esters are those derived from strong acids, e.g. hydrohalic or alkyl- or aryl-sulphonic acids, and may be made by the process of Specification 529,055. The primary or secondary amines may be aliphatic, aromatic or heterocyclic and include anilines, naphthylamines, aminopyridines, aminoquinolines, aminobenzthiozoles and piperidine. The products have therapeutic value. In examples (1) piperidino-N-acetpiperidine amidine is made from chloracetpiperidine amidine hydrochloride and piperidine, (2) 2-n-dibutylamino- and 2- laurylamino-methylimidazoline are made from 2-chloromethylimidazoline and n-dibutylamine or laurylamine, (3) 2-phenylaminoimidazoline hydrochloride is made from 2- chloromethylimidazoline hydrochloride and aniline, (4) 2-phenylaminopropyl-imidazoline is made from 2-chloropropylimidazoline and aniline, (5) 2- 11-naphthylaminomethyl)-imidazoline is made from 2-chloromethylimidazoline and α-naphthylamine, and (6) 2-(#:#-diethyl-#- phenylethylaminomethyl)-imidazoline is made from 2-chloromethylimidazoline and ##-diethyl- #-phenylethylamine. Other specified products are phenylaminoacetamidine, phenylaminoacetpiperidineamidine, phenylaminoacet-#-phenyl- -ethylamidine, 2- and 4-methoxyphenylamino-acetamidine, 2-(21-methoxyphenylaminomethyl)-imidazoline, 6-methoxyquinolyl-8- aminoacet- #-phenylethylamidine, 2-(6-methoxy - quinolyl-8-aminomethyl)-imidazoline, and 2- (6-methoxyquinolyl 8-aminopropyl)-imidazoline.
