An efficient one-pot synthesis of hydroxynaphthyl-substitutedglycine derivatives from 2- or 1-naphthol, glyoxylic acid, and benzylcarbamate via a modified Mannichreaction is described. The enantiomerswere successfully separated on analytical and semipreparative HPLCcolumns. Their absolute configurations were determined by CD analysissupported by TDDFT CD calculations.

A facile preparation of novel u-hydroxy-α-(2-hydroxy-1-naphthyl)acetic acid by a base-catalyzed condensation of 2-naphthol with glyoxylic acid is described. One of itsderivatives, α-methoxy-α-(2-methoxy-1-naphthyl)aceticacid, has successfully been resolved.
