An efficient route to acylated acyclic nucleosides containing a branched hemiaminal ether moiety is reported via three-component alkylation of N-heterocycle (purine nucleobase) with acetal (cyclic or acyclic, variously branched) and anhydride (preferentially acetic anhydride). The procedure employs cheap and easily available acetals, acetic anhydride, and trimethylsilyl trifluoromethanesulfonate (TMSOTf)

The reaction of substituted 1,3-oxathiolanes with diazocarbonyl compounds in the presence of Cu(OTf)2 and Rh2(OAc)4 was studied. The reaction is accompanied by the insertion of an alkoxycarbonylmethylene fragment into the C–S bond of the oxathiolane ring and leads to the formation of heterocyclic and heteroaliphatic products.

Purine and pyrimidine anions were alkylated using the iodomethyl ether and thioether generated by the reaction of 1,3-dioxolane and 1,3-oxathiolane with trimethylsilyl iodide.
