The synthesis of a fluorescein-derived fluorogenic substrate, D,L-myo-inositol l-([6′-hexyloxy-3-oxo]spiro[isobenzofuran-1(3H),9′-[9H]xanthen]-3′-yl hydrogen phosphate) sodium salt 9, for assay of activity of phosphatidylinositide-specific phospholipase C (PI-PLC), is described. This substrate is shown to be a sensitive substrate for bacterial PI-PLC in a continuous, non-radioactive assay.
