n-butyllithium to form the corresponding salts, M+[H(PO3R2)2], is described. These compounds were characterized by 31P and 1H NMR; their synthetic usefulness was demonstrated by the preparation of a variety of alkylates, R′CH(PO3R2)2, and by the preparation of mono and dihalo derivatives, XCH(PO3R2)2 and X2C(PO3R2)2, by direct halogenation. The first examples of alpha-azido bisphosphonates [(RO)(2) P(O)](2)CXN3 (1, R = i-Pr, X = Me; 2, R = i-Pr, X = H; 3, R = H, X = Me; 4, R = H, X = H) and corresponding beta,gamma-CXN3 dGTP (5-6) and alpha/beta-CXN3 dATP (7-8) analogues are described. The individual diastereomers of 7 (7a/b) were obtained by HPLC separation of the dADP synthetic precursor (14a/b).
