Heteroaryl and aliphatic analogs of diarylurea-based cannabinoid 1 receptor (CB1 R) allosteric modulators of formula (I) are described. Exemplary analogs can provide improved potencies and pharmacokinetic properties. Methods of using the analogs to treat diseases mediated by CB1 R, such as substance abuse and obesity are described.
A one-pot procedure was developed to prepare new 2-aryl-5-nitrothiophenes efficiently from bromonitromethane and 3-chloro-3-aryl-propenals. Nitrothiophenes were synthesized in good yields with a simple and easy workup procedure.

It has been shown that reactions 5‐substituted 2‐nitrothiophenes with arylacetonitriles in the presence of potassium hydroxide in methanol lead to the formation of the new [2‐(hydroxyimino)‐5‐R‐3(2H)‐thienylidene](aryl)acetonitriles. Under proposed reaction conditions, the formation of thieno[2,3‐c]isoxazole was not the case. J. Heterocyclic Chem., (2011).
