制备方法与用途

生物活性

RK-33是DDX3(RNA解旋酶)的小分子抑制剂,在DDX3过表达的细胞中,可引起细胞周期阻滞、诱导凋亡,并促进辐射敏化作用。

体外研究

RK-33特异性结合DDX3,而不结合DDX5和DDX17。它依赖于DDX3来抑制肿瘤生长并使肺癌细胞对放射线敏感。RK-33不对线粒体呼吸功能或ATP的产生造成影响,但能抑制增殖并诱导凋亡。此外,DDX3通过调节Wnt信号通路发挥作用,而RK-33则能够抑制这一通路。RK-33还通过抑制NHEJ(非同源末端连接)的活性来损害辐射诱导的DNA损伤修复。

体内研究

在多种肺癌模型的小鼠中,将放射与RK-33结合处理可诱导肿瘤消退。即使在所使用的浓度剂量下,RK-33对SCID小鼠也没有毒性作用,且经RK-33处理的小鼠没有明显的形态学变化。
