  Mandelonitrile lyase as present in a crude extract of almond flour has been tested for the synthesis of several chiral cyanohydrins. Silylated cyanohydrins of benzaldehyde, 4-methoxybenzaldehyde, piperonal, 5-methylfurfural, butyraldehyde and crotonaldehyde were obtained in good yield and high enantiomeric excess (93%) after treatment with silyl chlorides and imidazole in DMF.

  A new method for the preparation of enantiomerically pure N-benzyl nitrones is described. By using either a one-pot reduction-transimination or a one-pot Grignard addition-transimination sequence chiral O-protected alpha-hydroxynitriles can be converted into chiral aldo- and ketonitrones, respectively.   (-)-(R,E)-2-hydroxy-3-pentenenitrile 1, obtained by R-oxynitrilase catalyzed addition of HCN to 2-butenal, was shown to be an excellent chiral starting material for the synthesis of alpha-hydroxy-beta-amino acids of high enantiomeric and diastereomeric purity.
