    Provided are polymer compositions made by a process comprising: (a) providing a first reactive composition containing: (i) a polymerization initiator that is capable, upon a first activation, of forming two or more free radical groups, at least one of which is further activatable by subsequent activation; (ii) one or more ethylenically unsaturated compounds; and (iii) a crosslinker; (b) subjecting the first reactive composition to a first activation step such that the first reactive composition polymerizes therein to form a crosslinked substrate network containing a covalently bound activatable free radical initiator, (c) combining the crosslinked substrate network with a second reactive composition containing one or more ethylenically unsaturated compounds; and (d) activating the covalently bound activatable free radical initiator of the crosslinked substrate network such that the second reactive composition polymerizes therein with the crosslinked substrate network to form a grafted polymeric network and a byproduct polymer. Also provided are precursors to the polymer compositions, processes for preparation of the polymer compositions, and methods of using the polymer compositions, for instance in medical devices.

    A copolymer comprising at least one recurring structural unit of formula VII optionally at least one recurring structural unit of formula VIII and recurring structural units of formula IX wherein Rk is hydrogen or methyl, Rm is hydrogen or C1-C12alkyl, phenyl or benzyl, Rn ad Ro are each independently of the other C1-C12alkyl, phenyl or benzyl or Rn and Ro together are tetramethylene, pentamethylene, -(CH2)2-O-(CH2)2- or -(CH2)2-N(C1-C6alkyl)-(CH2)2-, and F is the radical of a fluorophore bonded directly or via a bridge group to the N atom, with the proviso that at least two different recurring structural units of formula VII or at least one structural unit of formula VII and at least one structural unit of formula VIII are present. The copolymers can be used for optical pH sensors.

    The FTO protein is unequivocally reported to play a critical role in human obesity and in the regulation of cellular levels of m(6)A modification, which makes FTO a significant and worthy subject of study. Here, we identified that fluorescein derivatives can selectively inhibit FTO demethylation, and the mechanisms behind these activities were elucidated after we determined the X-ray crystal structures of FTO/fluorescein and FTO/5-aminofluorescein. Furthermore, these inhibitors can also be applied to the direct labeling and enrichment of FTO protein combined with photoaffinity labeling assay.
